TERAPIA LOCAL PARA RESTAURAR LA AUDICIÓN Y TRATAR EL TINNITUS
Los resultados presentados aquí se obtuvieron en experi- mentos en animales, en los que los fármacos se aplicaron di- rectamente en la cóclea. Está claro que la aplicación sistémica de moléculas anti-apoptóticas o de antagonistas de NMDA causa efectos secundarios. Por ejemplo, los antagonistas de glutamato tienen efectos deletéreos en el aprendizaje y en la memoria cuando son aplicados sistémicamente. Similarmente, la aplicación sistémica de drogas anti-apoptóticas puede indu- cir la aparición de tumores. Por lo tanto, los avances en la far- macología local en humanos representan una gran esperanza para la conservación de la audición y el tratamiento del tinni- tus en pacientes expuestos al ruido o a las substancias ototóxi- cas.

El número de respuestas falso-positivas se muestra en función del tratamiento. En ausencia del sonido, los animales no ejecutaron la tarea durante un período silencioso (grupo control; n=10). En contraste, el tratamiento de salicilato (300 mg/kg/día durante 4 días) llevó a un aumento drástico en el número de respuestas falso-positivas, aún en animales que recibieron la aplicación intracoclear de perilinfa artificial (grupo AP; n=10). La aplicación local del antagonista NMDA 7 clorokinurenato (grupo 7- CK; n=10) abolió el aumento en el número de respuestas falso-positivas.
