La región IIa está constituida por los genes DQA, DQB, DRA y DRB. En bovinos se han identificado tres genes DRB; DRB1 (pseudogen), DRB2 (baja expresión) y DRB3; siendo este último el de mayor expresión y polimorfismo (Muggli y Stone, 1989; Burke et al., 1991).
BoLA-DRB3 es un gen que codifica la cadena de la molécula del MHC (Sitte et al., 2002)., su segundo exón (DRB3.2) codifica el sitio de unión del antígeno de las células presentadoras de antígeno; su alto grado de polimorfismo determina su capacidad de unión a un amplio rango de péptidos (Sharif et al., 1998; Russell et al., 2000).
En estudios recientes se han detectado delecciones o sustituciones de residuos en el sitio de unión al péptido BoLA-DR, que indican ser de importancia estructural en la definición del sitio de reconocimiento del antígeno (Sharif et al., 1998; Sharif et al., 2002; Xu et al., 1993).
BoLA DRB3 es el gen clase II mas ampliamente estudiado. Se han detectado más de 90 alelos diferentes (Takeshima et al., 200), los cuales potencialmente afectan muchos rasgos relacionados con inmunidad, SCS (Somatic Cell Score) e incidencia de mastitis (Starkenburg et al., 1997).
Dietz et al (1997) establecieron asociación significativa de alelos DRB3.2 con 13 rasgos indicadores de inmunidad innata y adaptativa, sugiriendo que DRB3.2 esta relacionado con factores de la función inmune y así, asociado con resistencia o susceptibilidad a enfermedades.
Aunque a partir de numerosos los estudios se han logrado establecer variantes moleculares asociadas a los diferentes fenotipos de interés económico en diversas razas y poblaciones (Heyen et al., 1999; Sharif et al., 1998; Xu et al., 1993;
extrapolados a otras razas o poblaciones debido a la variación en las tasas de recombinación ligada a la historia familiar (Thomsen et al., 2001).
A pesar de las limitantes tecnológicas y económicas en nuestro país, en la actualidad se están adelantando diversos estudios con los recursos genéticos criollos y colombianos, con el fin de determinar una luz en la búsqueda variantes genéticas asociadas a características de interés económico. En resultados iniciales de la caracterización del microsatélite DRB3 exón 2, en la raza lucerna, fueron identificados 24 alelos encontrando frecuencias alélicas desde 0.02 hasta 0.26; en este estudio se encontró una asociación significativa de uno de los alelos con un alto conteo de células somáticas y la incidencia de mastitis (p<0.05).
Existen numerosas alternativas para disminuir la incidencia de mastitis en una población, abordadas desde diferentes puntos de vista o de forma integral (nutrición, sanidad, manejo y genética animal). Así, el mejoramiento genético animal busca transformar a largo plazo, la incidencia de la enfermedad, mediante la selección de animales superiores genéticamente en condiciones adversas de producción. La selección asistida por marcadores genéticos es una herramienta de selección, que unida al mejoramiento genético tradicional, pretende identificar rápidamente animales superiores en las explotaciones.
